Q5 DNA polymerase : what it is?

Hello,

I was digging to find information on the Q5 DNA polymerase. I looked at the patents listed on the NEB product page and they are all expired. It seems that, like Phusion, its a polymerase with the Sso7d domain. But I couldn’t find the entire sequence.

Do someone have done some research on it and happen to have found the sequence fo Q5 ? It may be a Phusion with additional mutations.

Have a good day,

Louise

Hello Louise,

I don’t know if this will be much help, but kindly check the Google Sheets from Open BioEconomy Lab on DNA polymerases here. You will find that they both have the Sso7d domain. Hopefully someone can help with the requested sequence.

Best,
Franklin.

The sequence of Q5 is straightforward; what matters more is its PCR reaction buffer and its high-activity purification process

oh nice, can you share it with me ? Where did you found it ?

Thank you for your help !

I did have this information, both Phusion and Q5 are fusioned DNA plymerase with Ssod7 domain. NEB clearly mentioned than Phusion is Pfu DNA polymerase, but they say nothing about Q5.

From the Barrick Lab website, its mentionned that Phusion is a mutated version of Pfu-Ssod7 (https://barricklab.org/twiki/bin/view/Lab/ProtocolsReagentsPfuSso7d).

I also found this plasmid on Addgene where Richard Backer claimed is Phusion " Ssod7-fused Pfu Polymerase with fidelity enhancing mutation" https://www.addgene.org/228494/
