Molecular cloning remains central to synthetic biology and biotechnology and can occupy a substantial amount of training and working time, but there are few available software tools to support the management, documentation, and troubleshooting of DNA construction. CloneCoordinate is a free, open-source electronic laboratory notebook specifically designed to facilitate cloning at the individual, project team, or laboratory scale. It makes cloning robust toward failure of individual steps and accessible to participants with diverse experience levels. It provides a real-time prioritized task list, systematic sample inventory, and standardized data structures that enable collaborative and collectivized workflows. Crucially, CloneCoordinate captures the granular, specific data needed to assess and improve cloning efficiency and provide automated troubleshooting that can lower the barriers to successful cloning. CloneCoordinate could provide meaningful synergy with the Reclone project’s implementation and goals.
